Patented polymers, how do they perform lysis?
LNC(Lysis and capture) polymers chemically bind to fungal cell walls through charge difference, physically disrupts cell walls when vortexing is performed and biologically induces cell death.
At the same time extracted nucleic acids are captured by these polymers.
Due to their excellent performance heating is not required on lysis steps.

1. Q : What are the features of the PURE™ series that make it different from other extraction products?
A : Other products use pretreatment(shredder) columns and require two types of spin columns, resulting in a long or complicated extraction process. In addition, some products use organic solvents such as phenol and chloroform. The PURE™ series developed by Infusion Tech does not use pretreatment columns and enables high-purity, high-concentration nucleic acid purification with a single spin column. The precipitation process using organic solvents is also not required, making it a quick and easy three-step process (lysis, wash, elute).In addition, the lysis buffer of PURE™ Fungal gDNA Extraction Kit contains specially formulated polymer called LNC polymer.Unlike conventional silica membrane and magnetic bead methods, LNC polymers enable simultaneous lysis of fungal cell walls and capture of nucleic acids through charge difference.
2. Q : How do I use LNC polymer?
A : Since LNC polymer is a specially formulated polymer and has the properties of an insoluble powder, it is natural for precipitates to form. We recommend using the buffer immediately after vortexing, before precipitation occurs.
3. Q : How do I prepare a sample of mycelium?
A : This product is designed for fungi that are difficult to extract DNA from (Aspergillus sp., Rhizopus sp., Penicillium sp. etc.). After culturing the fungus, scrape off an appropriate amount(≤40 mg) of mycelium from the agar culture and add 100-200 μl of 1X PBS or distilled water before use. If it is difficult to weigh due to the large amount of mycelium, cut out together with agar culture, add 5~10ml of 1X PBS, perform centrifuge, measure the supernatant with a hemocytometer and use the appropriate amount(≥105).
4. Q : No quantitation is measured. Are subsequent experiments possible?
A : Various subsequent experiments including PCR and qPCR is possible. Since fungi species are mainly composed of mycelium and have very small amounts of DNA, they are very difficult to perform extraction. Also, it may be difficult to measure concentration with Nanodrop because the product contains a small amount of LNC polymer, which was specially formulated to boost lysis efficiency against fungi, during the elution process. Therefore, we recommend measuring concentration with a Qubit instrument or perform PCR rather than quantitation for accurate results.
5. Q : Are the components of the PURE™ Fungal gDNA Extraction Kit sterile?
A : The components of the PURE™ Fungal gDNA Extraction Kit are sterile.
6. Q : Which buffers in the PURE™ Gel Extraction Kit do I add ethanol to?
A : This product does not require ethanol. It can be extracted without direct addition of organic solvents. If you want to check for other components, please refer to the MSDS of the product.
7. Q : How is product quality control (QC) performed for the PURE™ Fungal gDNA Extraction Kit?
A : The QC of all products is performed by randomly selecting products of the same LOT after production and performing extraction using our standard sample. The test methods are quantification, electrophoresis, and PCR. Products are released if they meet our criteria.
8. Q : The filter was clogged while using it, why?
A : The amount of sample used to perform extraction is presumed to be high. We recommend re-experimenting within the sample volume range specified in the manual. If the amount of sample is small, it is likely that a low concentration will be measured when quantitating, so please reduce the amount of elution buffer.
9. Q : There are white powder on the wall of the spin column. Is there something wrong with the product?
A : The powder on the wall of the spin column is filter powder caused by static electricity during the manufacturing process. When the solution pass through the filter, the powder will be caught by the filter, and will not influence purity of the eluted RNA.
